The ideia of the pipeline was use a ensemble of initial conformation to start the design process, having multiple conformations of the interface to start the design process. using the pdb 7VSM provided, we first generated a ensemble for the interface of the complex using partial difusion. with the ensemble, we used the relax of rosetta on the structures and selected the top ones basing on the scores of dG separated and shape complementarity. after this, we kept the loop that is buried into the target and 4 sets of beta sheets, which 2 are close to the target and also a glicosilation. with this initial backbone, we generated the rest with rfdifusion, and filtered the best backbones using a score composed by radius of gyration + number of contacts with the targets. the top 20 backbones had it sequences designs (locking the initial motif sequence) and the best sequence for each one were used as final candidate.
