Loops interacting with the NIPA proteins were extended to increase contact area between the binder and the target protein. Loop structure candidates were made using RFdiffusion, sequence was designed by ProteinMPNN, structure validation and ipSAE_min scoring was performed using both AlphaFold2 and Boltz2.
