The Latent Labs binder designs were generated with a fixed length of 80 amino acids (after testing multiple lengths), and the hot spot residues used for targeting were those indicated in the image: 494, 496, 497, 498, 500, 521, 522, 523, 547, 549, 550, 572, 573, 574, and 575. As the flexibility of the Latent Labs platform is quite rigid in fine-tuning parameters, the core rationale about the design comes mostly from literature review of residues involved in Nipah binding with partners. The 5 top-ranked binders were selected based on optimized on visual inspection of the predicted complexes as well as iPTM and iPAE metrics.
