BoltzGen is an all-atom diffusion-based generative model developed for de novo design of protein and peptide binders across a wide array of target types. Differing from earlier approaches, BoltzGen unifies protein design with high-accuracy structure prediction, allowing for simultaneous generation of functional sequences and their three-dimensional atomic structures. The model supports a powerful design specification language, enabling fine control over covalent bonds, structural and sequence constraints, specific binding sites, and more. The BoltzGen design pipeline has achieved state-of-the-art binding and folding performance and has been experimentally validated in diverse wetlab campaigns across dozens of challenging biological targets, including both proteins and small molecules.
