We designed de novo minibinders against Nipah virus Glycoprotein G head domain using FoldCraft or BindCraft. To make design process faster we trimmed the head domain of Glycoprotein - 2VSM (337-511). As the trimming exposed the big hydrophobic area, where all FoldCraft (BindCraft) designs were drived, we redesigned the core region of trimmed Glycoprotein with SolubleMPNN to create a small highly soluble protein particle. We then designed de novo proteins with FoldCraft (or BindCraft) until we reached 100 designs passing BindCraft filters. We then repredicted all designs with Boltz2, and selected top10 performers by ipSAE metric.
