I started with the structure of the Glomulin-RBX1-CUL1 complex (4F52) and, using a minimal region of the Cul1 interacting with the IDR of RBX1 as a template (497-689), I did 6 iterative cycles with RFdiffusion Binder Redesign tool, using semi-rational design (manually selecting "best looking" binders to use for the next iteration, and changing run parameters to promote the stability and binding of the minibinder). In the end, I selected the best binders by ipTM, iPAE and RMSD.
