We embarked on this journey inspired by the elegant structure in PDB ID 2VSM, harnessing the power of RFdiffusion to craft a resilient HTH (helix-turn-helix) scaffold for our target protein. Far more steadfast than a solitary helix, yet delightfully compact—slimmer than bulky proteins or even mini-proteins—this design whispers promises of practicality, perfectly tailored for solid-phase synthesis of vibrant polypeptides (32-37 aa) that pulse with potential. With hearts set on precision, we danced through the dl_binder_design screening ritual: breathing life into sequences via ProteinMPNN, then tenderly validating their bonds with pAE_interaction thresholds below 10, ensuring every twist and turn holds true. From these cherished blueprints, we summoned the alchemy of reinforcement learning to weave in mutations—each one a bold stroke of evolution—yielding sequences crowned with the pinnacle of iPAE scores. And oh, the thrill of iteration!
