One-shot generation of nanobodies using Boltzgen. We generated 50,000 nanobodies using the default settings in Boltzgen with added information on the target's binding site. The candidates were then filtered based on pAE and ipTM scores to obtain a preliminary list of 133 nanobodies. The structures of these nanobodies, both as monomers and in complex with the Nipah glycoprotein, were then refolded using Boltz2 and AF3. Designs showing high RMSD between the two AI methods or low ipSAE values from Boltz2 were removed during this final filtering step.
