Description
Our design and selection workflow used ODesign for candidate generation, and final scoring was based only on:
- Rosetta dG (binding energy-related score)
- AlphaFold 3 ipTM (interface confidence score)
For each candidate, we calculated a combined score using equal weights:
$$ S_{combined} = 0.5 \times S_{dG} + 0.5 \times S_{ipTM} $$
where $S_{dG}$ and $S_{ipTM}$ are normalized scores derived from Rosetta dG and AF3 ipTM, respectively.
Candidates were ranked by this combined score, and the top 5% were selected as final high-priority designs.
