Pipeline config (V3): 2,500 de novo candidates; heptad-constrained helices with hydrophobic core placement, surface charge balancing, Cys<=1, Pro<=1, charge fraction 25-45% (DEKR), low-complexity and hydrophobic-run filtering, internal diversity filter (>80% identity rejected). Top 200 candidates underwent Boltz-2 ensemble scoring across 4 RBX-1 conformations (800 total predictions). Final ranking used six weighted signals: mean ipTM (0.30), mean confidence (0.10), inverted mean iPDE (0.10), physics regularization (0.20), first-pass foldability (0.15), ESM-2 PLL per-residue (0.15). Submission composition: top 30 unique Boltz-validated cores + 70 single-residue apex variants. Sequence length range: 79-94 aa.
