The sequences in this collection were created using Cradle’s zero-shot diversification pipeline, which introduces targeted mutations around a functional lead to explore nearby sequence space. Starting from an scFv-formatted Cetuximab, Cradle diversified the framework region while keeping the CDRs intact, producing 12 variants that all showed strong binding to EGFR. One design achieved a 1.21 nM affinity, eight times stronger than Cetuximab, and several others reached subnanomolar binding. This dataset includes all 12 designed sequences along with their experimental binding data and computational analyses.
